Overview

Biomedical research utilising animal and tissue models requires histological analysis as a key methodological process. The practical application and theoretical underpinnings of tissue histology is therefore an essential skill for all biomedical scientists. The purpose of this unit is to provide you with the knowledge and understanding of the application of histological techniques routinely used in research laboratories. This unit also provides an opportunity to develop practical skills in a range of histological techniques including tissue sectioning (microtomy), common histochemical stains and immunohistochemistry. The unit is positioned in the advanced phase of the course and builds upon the introductory tissue concepts covered in Human Systematic Anatomy (LQB183) and concepts of tissue adaptation in Developmental Anatomy & Tissue Adaptation (LQB382).

Learning Outcomes

On successful completion of this unit you will be able to:

  1. Demonstrate a theoretical knowledge of the technical details and applications of histological techniques and associated microscopic methods used in biomedical research.
  2. Demonstrate an ability to critically analyse the quality of stained tissue sections, including an ability to trouble-shoot technical problems and prescribe solutions for their correction.
  3. Demonstrate practical skills in histological techniques including tissue embedding, sectioning and staining, whilst exhibiting appropriate laboratory health and safety behaviour.

Content

The content of this unit includes:

The theoretical background and practical application of techniques used in the preparation (fixation, processing, embedding), sectioning (microtomy) and staining of research tissue samples. Coverage of basic and advanced microscopic techniques used in the analysis of stained tissue samples. An introduction to methods of data analysis used by research histologists.

Learning Approaches

You will be exposed to a variety of learning situations including online teaching materials and practical sessions. Practical classes will be held in the Gardens Point Histology laboratory (Q711) which gives you access to industry-standard equipment and chemical reagents. You will particularly develop technical proficiency in sectioning and staining of paraffin embedded tissue samples, and laboratory time management. Self-directed learning will be encouraged by giving you access to on-line resources and exercises in your accompanying teaching and learning manual. Feedback on your progress will be provided through regular lecturer-led, peer-group discussions of student work aided through use of a multi-user online microscope-based tutorial. The practical environment also provides an opportunity for ad-hoc discussions regarding opportunities for research-based higher degree projects and career development.

Feedback on Learning and Assessment

Throughout the semester you will receive immediate feedback on your laboratory work during practical sessions. Images and other materials that are available on Canvas are designed to consolidate your knowledge and give you immediate feedback on your learning. A take-home progress theory examination is held and feedback is provided in class. During semester and at the end of semester you will also be provided with the opportunity to give your feedback on the teaching and content of this unit. Your feedback and suggestions will be used to make changes to the unit where possible to improve the learning outcomes for you and future students.

Assessment

Overview

Participation in the practical classes is an integral part of your learning experience in LQB671. If you are unable to attend a practical session, you should inform the unit coordinator by email and provide supporting documentation for your absence the following week at the start of practical class, e.g. medical certificate. For approved absences alternate arrangements will be accommodated on up to two occasions. Approved absences in excess of two occasions will need to be discussed with the unit coordinator.

Assessment Tasks

Assessment: Tissue and Stain Identification exercise

This Timed Online Assessment will test the ability of students to identity different tissues and stains, based upon their interpretation of a series of high-quality digital images of histological specimens.

Assessment: Staining exam

This practical examination will require students to plan and subsequently execute a panel of different staining techniques within a designated time period.

Assessment: Final Theory Exam

This examination will test your knowledge of basic terms and definitions relating to common histological techniques. You will also be examined on your ability to apply this knowledge to the interpretation of stated staining outcomes and to be able to discuss potential solutions to common technical problems. You will also be required to evaluate the value of histological data from a research perspective and to discuss alternative presentation strategies.

Safety and protective equipment

Practicals will require personal protective equipment supplied by each student:
1. Laboratory Coat - 100% cotton or polyester/cotton mix
2. Safety eye protection glasses - selected and used in accordance with Australian Safety Standards
3. Fully enclosed non-slip shoes

Risk Assessment Statement

This unit includes a laboratory component. In order that you become proficient in necessary practical skills, you will be trained in the handling of equipment, materials and specimens normally associated with this discipline. You are required to view a Health & Safety Induction video and complete an online health and safety quiz prior to being permitted into the laboratory, to access please follow this link Health & Safety Induction. A passing score of 10/10 is required and will be checked prior to entry to the first laboratory session. You may repeat the quiz until this score is achieved. You are also referred to the Faculty of Health's Health & Safety Web Site for further information.

If you have an underlying health condition you are encouraged to consult the lecturer in charge of the unit, as you may be required to take additional precautions on occasions.
